Cancer immunotherapy has heralded a transformative shift in oncology, leveraging the immune system to achieve durable responses across diverse malignancies. Recent innovations—ranging from checkpoint blockade and CAR-T cell therapies to personalized RNA vaccines, oncolytic viruses, and novel delivery platforms—are extending therapeutic reach into solid tumors and refractory disease. This review synthesizes cutting-edge developments from 2024–2025, emphasizing mechanistic insights, clinical trial outcomes, and delivery innovations. Key challenges—immune evasion, resistance, toxicity, and logistical constraints—are addressed alongside emerging solutions like nanocarriers, biomaterials, and computational multi-omics. This synthesis highlights a new era in immuno-oncology that bridges bench and bedside.
